血液抹片染色 (hematological stains)

這個禮拜三Group meeting的報告是負責介紹Staining

包含Stain history, Wright’s stain, Giemsa stain, Liu’s stain, PAS stain (bonus)

 

上個禮拜做了很多片的Liu’s stain & PAS stain

查了資料之後，這下對Staining的了解又更多了:)

 

Liu’s stain

Procedure

1. Cover the slide with 5~6 drops of Liu A. Wait 30 seconds.
2. Add 2 times of Liu B on the slide. Mix well by blowing the surface. Wait 60 seconds.
3. Wash off the staining solution with running tap water(at the back side).
4. Dry and cover

 

PAS stain

1. Procedure
2. Warm the Schiff’s reagent to room temperature
3. De-wax and hydrate paraffin sections
4. Oxidize for 10-30 min in periodic acid
5. Wash in running tap water for 3 min
6. Immerse in Schiff’s reagent for 20 min
7. Wash in running tap water for 10 min
8. Apply counterstains, as desired ( hematoxylin, 90 sec)
9. Dehydration in alcohol
10. Clear in xylene and cover
